I saw the third wave of the gelatin guys come in, but I can't find them in my colosseum. Do you have to unlock it? or were they all used already?

You don't need to unlock any of them. Looks like they're all gone, I was fighting them earlier and there only a few hundred left. They should be back, though, at some point.
